Yes. Telling people their zipper is undone is one of the precious joys of life.
Don't restrict this activity to only the times that the zipper is actually un done. Mix it up a little.
Get others to play along. Women at work miking snide comments about immodesty prior to the prank can multiply the effectiveness and include the whole room in the enjoyment.
The best time to play is when he is making a presentation to a full room. Make sure several in the front row are in on it. As trey glance down the and get him self conscious.
You then stand at the back of the room and signal that the cow is out of the barn. It works every time.

If your zip is undone, could you like your colleague to tell you? Apply the same principle as what you want others to interact with you.

As a question of etiquette yes you should tell him in private that his zipper is down... or as my fiance` says to our 2 boys "Hey there zip!" and then points down when they don't get it.
OK that's not nice, but neither is having your zipper down all day ... what if he has to do a presentation?
