There are a few ways to tell....one, her heat cycle will stop, two her nipples will get bigger and raise. ( they will become alot more noticeable than before she was pregnant ) She may also become sensitive to anyone touching her belly. The average pregnancy lasts 63 days so if she is pregnant it wont be that long before she has puppies.

First, she has to be old enough to get pregnant. Dogs can have their first heat (and pregnancy) any time after they are 6 months old. Then, she will have to have mated with a viable male dog.

If she's old enough to get pregnant and has been bred (whether on purpose or by accident), she will start showing her pregnancy at about 40-45 days into her 68-day pregnancy. Her belly will start to swell, her nipples will get bigger and may "pink up". She may vomit occasionally (the doggie version of morning sickness); she may be off her food as the growing pups push against her stomach, and may have to urinate more often as the pups push against her bladder.

If she's "showing", you can take her to the vet and ask for a sonagram or xray to see if she's truly pregnant and how many pups she may be carrying.

(Keep in mind that some dogs have "false" pregnancies, so going to the vet helps to verify everything.)
