Probably not a good idea because olive oil has a much stronger flavor than vegetable oil and it might make the cake taste funny. Strangely enough, a substitute that does work is using unsweetened applesauce instead of oil with boxed cake mixes. You just swap the same amount of applesauce for oil. I do this all the time and it works.

YES!
I was just making a cake and wondered the same thing since I only had olive oil in my kitchen!
I researched it and found out that it would be OKAY to do--since baking with it tend to make the flavor of the olive oil diminish and you don't even notice it.
I just made a cake with it and it turned out FINE and was really good too!
Here are some links that talk about olive oil and specifically about using it to make cakes and baked goods too!

Use Canola oil , Walnut Oil, Almond oil etc. Olive Oil has its own taste, which is not noticeable in salads, when garlic and other strong spices are added. I personally do not like to cook with it at all, even heating the Olive oil , gives the house an odor I do not care to live with. If you are used to the odor of Olive Oil, and like it , let me not stop you from trying it in the Cake, you may even like it.

I do all the time, but I generally use a tablespoon or so less of olive oil. I never use vegetable oil in food preparation as olive oil or canola oil is healthier.

The olive oil would be too strong for your cake mix.

No! That's not a flavor you want to add to a cake. But you can use canola oil.
