Yes, you can do it at home. It's not hard, but you want to wear rubber gloves.
Here is a site that tell you how.

Most dogs drain their glands normally when they eliminate. You can increase the amount of fiber in her diet to make her eliminations more forceful to drain the glands.
Some dogs do get impacted anal glands which is usually the problem when they scoot their hind ends on the floor or lick at their rears. Impacted glands can be painful and can become infected. In rare cases, some dogs have them removed.
Some dogs also need more than one draining so it makes sense to learn how to do them if your dog has this problem. It can take some squeezing and it is unpleasant and your dog probably will not be happy about it.
Just make sure that your dog needs your help before doing it. Even if after seeing the pictures on the web you are unsure how to do this, you should ask your vet to show you how if he or she thinks it is even necessary.

If it's the anal glands you're talking about, take it to the groomer, if not the vet, and request that they do it. You can even request that they show you how to do it. This used to be common practice in grooming, but now they require that you specifically ask for them to do it if you want. Make sure that you call ahead of time to ensure a trained groomer will be doing it. If done at the groomer, it gives you the added benefit of getting your dog a bath and haircut at the same time =)

This is a nasty job, but yes you can do it at home. When you are holding your dog, do not hold his rear end facing you, but rather get to the side. Put your finger at 5 and 7:00 on his glands and squeeze. Do not squeeze to hard as this may hurt your dog. If nothing comes out, they may be impacted and needs a vet's attention. After that, you can do it yourself every couple of weeks or so.

Absolutely you can do that at home. However, I would go to the vet and let him do the first draining so that it is done properly and then ask him to show you so that you can continue doing it at home when needed.

You can.. but you might want to get your vet or groomer to teach you how to do it.. Might also wish you had paid the couple of bucks and let someone else do it.. You ever smell that stuff???
Good quality food, lots of water, and good exercise and you shouldn't have much problems with the anal glands.

You can, but you'll want your vet to show you how. I think you might change your mind about doing it yourself after the vet shows you how. Its NASTY.
