My boyfriend has a class B driver's license in NY, but has expired. Does this mean he has to retake everything all over again here in CA?

He MAY be able to simply transfer his class B to a California class B. But other factors may come to play, for instance, if he has a passenger endorsement or air brake or hazardous material endorsement.
These require a separate test and may also be true for your boyfriends'.
I recommend doing a search for California Department of Motor Vehicles. The website is extensive and has a fairly comprehensive FAQ page.

If his license has expired, he will probably need to take the CA test(s).
