The microphone input of my computer doesn't work when i plug the guitar amplifier from "headphones" out, instead it does when i plug it from "speakers". Why?

The headphone jack has higher impedance (1000 ohms or so). The speaker one is usually low (4 ohms or so). Go with the one that is a better impedance match to be able to transfer more energy.

Try reversing your wire in/out portals.
