Yup, Metro is the best way to get around - no need for a car.
Last time I went, I got to see all that I wanted to in one day. I just walked up and down the National Mall, starting at the Lincoln Memorial, then WWII, Vietnam and Korean Memorials, then Wash. Monument, then the Smithsonian Museums, then Walked up to the Capital building and White Houses, but didn't take tours of either.
2 days should be enough for you to see the most important things on your list, but certainly not everything. You'd need a month to see it ALL.

The Metrorail is a cheap, effective way to get around. I used it once on my way to a tour of the Pentagon.
I don't think you can visit everything in two days. In three days, I went to:
National Cathedral
National Archives
U.S. Capitol (tour)
Union Station
Jefferson Monument
Franklin D. Roosevelt Memorial
Washington Nationals Game
Lincoln Memorial
Vietnam Veterans Memorial
Korean War Memorial
White House (tour)
Ronald Reagan Food Court (got on Metrorail)
Pentagon (tour)
U.S. Air Force Memorial
U.S. Marine Corps Memorial (Iwo Jima)
Arlington National Cemetery
Mt. Vernon
Holocaust Memorial Museum
Washington Monument (went to top)
World War II Memorial
Old Post Office
Ford's Theatre
I also went to Baltimore, Maryland, and Gettysburg, Pennsylvania, but I didn't include them.

This is not the best or cheapest way, but a suggestion nonetheless. If you have money to burn, you can spend $28/each and do the Old Town Trolley ride which runs between 9-5pm and takes you to all the "touristy" stops. You can hop on off between those hours and get a mini history lesson while you're there. It also takes you to Arlington Cemetery & Iwo Jima.

The Metro subway system will take you within a few blocks of whatever you want to go to in DC proper, and some places in VA, too. Metrobus is the bus system.

a car would be useless. get the All day Metro Pass. after 0930 on the week days and all day on the weekends. unlimited usage of the Metro. you can get on on and off as many stops as you like on all the lines for one low price. I know for us it is actually cheaper to buy an all day pass when we travel from the Greenbelt station to the Zoo.
Hint: DO NOT get off at the Zoo stop as marked on the Red line. go one further to Cleveland Park. it's level as opposed to being uphill and just about the same distance.
There are also unlimited bus passes you can purchase as well.

No you can't see everything in 2 days. Impossible!
Do not get a rental car! Parking is difficult and you will get lost!
