I'm a native New Yorker.

I would start at South Ferry.

Take the Staten Island Ferry back and forth to Staten Island (it's free) and be outside. It's a beautiful trip. You will go very close to the Statue of Liberty, so have your cameras. It's a half an hour each way.

Get off the ferry. Walk along the Hudson river north (thru Battery Park) and you can walk past the World Trade Center site. Cut over (you'll need to ask directions, too complicated for here) to Fulton Street, walk toward the Seaport, turn Left on Broadway, bear right at the fork and go the City Hall train Station. Take the #6 to Canal Street. Get off. You will be in Chinatown.

Walk to Mulberry Street. Turn left. You will be in Little Italy. Try Ferrara's for dessert.

Then walk up Mulberry and you will go through Soho and then to the East Village. Go to Bleecker Street. Take the 6 train again to 33rd Street. Get off and you will be able to walk to the Empire State Building. 1 hour.

I am guesstimating that your tour will take about 5 hours or longer if you walk slow or linger in places.

Avoid long lines so stay away from the Empire State Building and the Statue of Liberty. Go to Canal Street in Chinatown. Then to Little Italy on Mulberry Street and start heading north to Soho. You see plenty of interesting art galleries and shops there to keep you busy for hours. And if you want you could keep walking north to Washington Square Park in Greenwich Village.

Grey Lines offers a double decker bus tour that will take you all around the city. The upper West side, Harlem, all the way down into the financial district, up and back on both sides of Central Park. More than you could possibly see in such a short time walking. I believe they offer a jump on and jump off option, that lets you get out for a while at a particular spot.

Take Poet61's tour. It's exactly right. I hope everyone in your group likes walking since New York is a walking city and you will do a lot of it. After the show walk around Times Square. It's spectacular at night and something that will top off a great day in New York.
