I'd honestly say down by the river. I think it's a Hyatt right there on Columbus Blvd. (Delaware Ave.)...it's right on the Delaware River, and right across the street from Old City and South Street. Even though traffic gets busy on that road, it's a river view and there are plenty of things to do in Penn's Landing by itself without ever even having to cross the street.
Even better is Society Hill. There's a Sheraton right there that's located within walking distance of South Street, Old City, and most of Philly's history. South Street isn't too bad near Headhouse Square (closest to Society Hill) but as you go further west, it gets kind of young (I guess?) and becomes more music stores than bars and restaurants. Old City is a little bit further away (a nice walk though) and is mostly upscale dining and clubs. Then there's the Liberty Bell, Mint, Constitution Center and Franklin Park.
There's obviously so much more to see and do, but I'd have to say that's an ideal spot to start out at.

I agree with everyone else. You really can't go wrong in center city, which is roughly the area between 1st and 20th streets, south of Vine street and north of South street (got that ?I). You're close to a lot of stuff, including the theater district, old city (independence hall, liberty bell) and LOTS of nightlife and eateries. You'd also be close to the subway (underneath broad st, runs to the sports stadiums) and the septa station at 10th and Filmore (takes you to and from the airport & suburbs/megamalls)

Center City or Old City would be best. This way you would have easy access to great restaurants, bars, clubs, shopping and points of interest all within a reasonable walking distance. If it is not withing reasonable walking distance, you would be able to easily take advantage of the SEPTA system to get you wherever you needed to go. Enjoy and don't forget to try a cheesesteak!!!

Center City has a mix of standard hotels (Holiday Inn) and upscale hotels (4 Seasons) so you have a lot of choices. It is walking distance to Market street shopping and to Chinatown and South street and to theatres. Loads of restaurants and also accessible to historic district.
