Driving you can take the LIE to exit 62. Then take Nicolls Road north to Stony Brook. It is about a 45 minute drive.
For train or bus you need to look up their schedules.

From Wantagh, the most direct way would be to get on the Wantagh parkway northbound to its end where it connects with the Northern State parkway, then go eastbound on Northern State all the way to the end where it joins up with highway 347. Take 347 east all the way to Nicolls road and turn left to get to SUNY Stony Brook.

There is no direct rail line from Wantagh to Stony Brook. It's not a good idea to take the train because you will need to transfer 3 or 4 times between different lines of the LIRR to Ronk, and then you have to take a bus.

Taking the bus exclusively would be tedious as well because you got to take Long Island Bus in Nassau into Suffolk County, then transfer over to Suffolk County Transit Bus.

I hate to say it since I'm an advocate of mass transit.. But your best option for this trip is to drive. :-/
