Depends on where you are flying to. When you check in at the counter where your flight originates, ask if your luggage will be sent right through, and they will inform you if you have to pick it up. In some places you pick up your luggage and put it on another luggage belt your self. Most of the time though it is sent straight through to your destination.

If you're changing airlines on your connection (called offline travel) , and the two airlines do NOT have a baggage agreement, then you have to claim your luggage and recheck-it. Jet Blue for instance has no baggage agreements with anyone but themselves. Same goes for Southwest.
But two airlines that do have an agreement, plus all online travel, ie two flights in the connection on the same carrier, or same coded carrier, like Delta to Delta Connection, Jet Blue to another Jet Blue flight etc, your bags are checked through to your final destination.

If you're connecting to an international flight, ie flying New York to Los Angeles rthen connecting to Sydney Australia, and your carriers have a baggage agreement, then your bags are checked through to Sydney and you don't have to claim/re-check them.

Only when arriving at an international city and then immediately connecting onward do you *usually* have to claim your bags in order to clear customs/immigration then recheck them for the new flight, though in many cases you just have to carry them out of immigration and place them on a conveyor, the bags are pre-tagged with the final destination.

Only if your changing airlines, if your staying on the same airline they will do it for you. The only exception is when you fly international you have to get yours bags and recheck them after you go through customs.

But if your flying domestic, same airline than nope.

If it's an international flight, you do. If it's domestic, you check the bags through all the way to the end.
