If you're driving it takes about 4 hours from Florence to Naples (489 km) and then roughly another 2 hours to Amalfi.
By train it takes about 3 hours 20 minutes (high speed trains) from Florence to Naples, then you have a choice of ferry, bus or taking a train to Sorrento (circumvesuviana trains) and then a SITA bus. The circumvesuviana trains run about every 30 minutes or you can take the SITA bus from Naples to Amalfi.
You could also take a train from Florence to Salerno (the best connection is changing in Rome, but there are other options; 4 hr 23 min). In Salerno the SITA bus, runs along the Amalfi coast, or there are ferries that are faster. The boats are about 10 minutes from the train station.

Depends where along the Amalfi Coast you want to go. You might want to go to yahoo maps and type in the locations you want to travel from. You'll get approximate distances and travel times.
