Some do require you to uninstall them while others do not. I err on the side of safety and always uninstall the older version first before installing the update. It's painless and can save you a lot of grief. Certain programs can cause serious damage if they are not uninstalled first before updating. For example, many security software (e.g. Norton Antivirus) and multimedia packages (e.g. Nero) require you to uninstall previous versions.

Depends on the software - check the readme file or installation guide. Some like Norton Antivirus requires you to uninstall first or things can get really messy. Sometimes, even a program update may request a removal first or it will remove the entire software suite for you! That happens for NTI CD and DVD-Maker.
