There are no calories in a pound. A calorie is a unit of heat energy, a pound is a unit of mass.
Ohhhhhhh... I bet you want to know how many calories you have to burn to lose a pound of fat, huh?
"...there are 3500 calories in 1 lb. of body fat..."

1. Burn the substance in a Bomb Calorimeter. An open cup version may also work for a larger quantity, but precision suffers.
2. Measure the heat released by the small sample by measuring the change in temperature surrounding the Calorimeter.
3. Calculate and convert
Q=c(Tf-Ti)*Ms
Q(M)=Q*M/Ms
Q=heat, convert to calories depending on your other measurements.
c=specific heat capacity of bath
Ms=mass of sample
M= mass desired
