Any walking, running or stair climbing will give you alot of muscle. But, climbing stairs is not the best way to do it because you have 50% "down time" when you run down the stairs (which is very low impact and has almost no positive effect on your muscles).
A stair machine at your local gym is your best way, walking is the runner-up

Of course it will build muscle between your calfs and butt. You can switch up the way you do the stairs between using every step or every other step. Or you can step on the edge of the step and push off to feel the burn in your calf. Run backwards down the stairs to reverse the muscle.
Don't forget to stretch also, it'll help tone and build muscle.

Stairs will help tone/build your buns and your upper thighs primarily. There's some cardio there too as it takes more effort to lift your body up a flight of stairs than to carry it across a flat surface.
I don't know that it's great as your only method of exercise, but it beats the heck out of taking the elevator and is one of the reasons there are stair-climbing workout machines.

i was a runner for about 8 years and yes it will build muscle in your butt mainly and since you will be on your toes it will build calf muscles. it works the thigh muscles some but not as much as the buttox and calves. every work out you do will work different muscles. my legs were rather big so it doesn't make them skinnier at least not in my case. they were big from muscle though not flab.

I guess ifyou do it for long enough. I would go the thealth club though, they have a machine that is like a stair treadmil the stairs just keep rolling down on you.
