Don't delete anything if you don't know what it does. Couple of things to speed up your PC. Let Windows do a clean up using System Tools located under accessories on your program Start Menu. This will clean the temp internet files as well as the temp files.
Uninstall programs that you don't use.
Keep your computer clean using an anti spyware program and an anti virus program
Defrag on a regular basis

All the above answers are entirely accurate, but Microsoft has an easy web-based program which can do all those things.

Empty your recycle bin weekly. Delete temporary files (In Internet Explorer: Tools>Internet Options>Press "Delete cookies" and then "Delete files".) Don't touch system files or anything that's in C:\Windows.
