Towards the end of the film, when he realises that life is worth living and that he has the love of his friends and family and realises thats all that matters. You laugh and cry all at the same time.

The scene in S.O.B. when they give Felix Farmer a Viking funeral at sea. It's sad and funny at the same time, because the camera alternates between shots of the flaming row boat with Felix's body drifting off into the distance, and the location, miles away, where his widow, Sally Miles, is singing at his memorial service beside a casket that, unknown to anyone at that service, actually contains someone else's body. Felix's buddies had gotten drunk the night before and decided he deserved something better than the big production all his fair-weather friends were putting on.

The scene in Election where Jim McCallister sees Tracey Flick again...it's so infantile and funny, but it's great the the character gets to have his catharsis.
Other favourites include the chase scene in Ronin and the scene with Patrick Bateman dancing to Hip to be Square in American Psycho.

I have two:
From "The Godfather": The baptism of Connie's baby with Michael verbally renouncing Satan and all his works - juxtaposed against images of Michael's henchmen, at Michael's orders, assassinating Michael's rival, underworld bosses.
From "Patton": During the Battle of the Bulge, Patton (in voice over) reads the "good weather" prayer the chaplain has written for him while images of the carnage of war are splashed across the screen. As the prayer ends, a lone soldier in silhouette walking across a snow covered field gets shot in the back and falls dead onto the hard, cold ground.

OK, I have a few:
LOTR
1- Baromir saving Merry and Pippin (true valor, even though the guy tried to take the ring) 2 - After Arwen returns to her father and tells him that he saw her son in the future: Arwen drops her book and she says "this was my choice." --- uhhh.. grips you.
A Walk to Remember: 'I don't need a reason to be made at God.'- Mandy Moore to Shane West. And the ending scene of Shane West walking on the dock 'feeling the wind.'
Anna and the King : As the king comes to Anna who is listening to the music box--she cries as she explains that science can explain the way of music, but is unable to explain the relationship with her and a king. They dance with the realization that they truly do love each other, but are unable to continue in that relationship. (I did not cry in this part, but man, is does it take a hold of you and make you think---are you strong enough to do what is right?)
Pride and Prejudice- As Mr.Darcy leaves Lizzy with the letter explaining the charges she brought against him.
hmmm...there are so many.
I must say that I really enjoy the scenes that are most gripping and truly challenge you as a person. -- They can tend to be melancholy moments. The character really exemplifies who he or she truly is.

There was this film I've got at home called 'The Gods Must Be Crazy' and in it there is a classic scene with a guy getting chased by an ostrich!!!!!!
Its done at double speed as well so every single time I watch it I crack up with laughter! :)

one of my favorite scenes is from true romance when patricia arquette is in the car with the movie directors assistant , they get pulled for speeding and he is trying to hide the bag of cocain when it bursts covering hi in white powder as the policeman walks up to the car , the first time i saw that scene i nearly wet my self with laughing

Two scenes from The Color Purple, When God is trying to tell you something and the daughter hugs her dad. I cry every time. and the end, when the children come home, boohooing. Love it.
I also loved in 300 when the wife killed her rapist!
