Based on my lifelong residency and working life in NYC, I can safely say SoHo stands for the area South of Houston Street. The area is North of Spring Street and West of the Bowery, the section that is NOT part of Little Italy or Chinatown.

SoHo is a shortened version of South of Houston street, which is where the area is located. Another New York neighborhood, TriBeCa, stands for the Triangle Below Canal Street and NoHo is...yup, North of Houston street.

South of Houston. And houston is a highway that runs east and west. It fantastically plenty separates the quantity streets (working east and west) from the worded highway names (additionally working east and west).
