There are these long lists of instructions called Check Lists.
Each Astronaut gets to work on one group of check lists which are focused upon his special assignment and training.
they go through these lists all the time looking for aberations, and green versus red lights. Other time is spent eating, sleeping, doing exercise and conversing with the control center back on Earth. Some specific scientific experiments are conducted according to a preplanned schedule, and various types of repair and upgrade activities are routinely performed. Each man has a specific set of duties and normally takes care of those functions. Some overlap and cross training is provided in the event that one or more of the crew is ill or out of action. A rigorous set of medical examinations are also performed on a constant basis using data links with the ground stations.

Firstly we haven't been to the moon in like 38 years, but then they took samples, examined the terrain, put up a mirror and flag, put up seismograph, among othere things.
On the international space station, they just do expiraments that can't be done on earth, and they make observations that aren't distorted by the atmosphere.
On the shuttles, the repair many sattilites, bring parts of the ISS, and bring more crew. Again with the expiraments.

If they aren't busy maintaining and building the space station they are doing experiments ... or sleeping ... not much spare time on a space mission.
