yes. most duel battery setups use the primary battery for the starter and the secondary battery for other things. In, fact it the batteries do not have an Isolater between them, I'd disconnect the second battery till you have it started. Primary is usually the one on the passenger side of most vehicles. To ascertain if it is, follow the red cable, it should go to the starter solinoid. If so, that's the one to jump to. Also, be sure you only hook up the red cable to the positive post on both cars, but the black negative cable goes from the negative post on the running vehicle, to a solid ground on the dead one. Not to the negative post.

Chances are the vehicle is like mine, I have 2 batteries because I need the extra CCA to get my truck started sometimes. Anyway, it does matter, always jump the battery that is connected to the starter, typically the one on the passenger side.

I've never seen a car with two batteries. Seen plenty of trucks with two or more batteries. But we both know a car and a truck aren't the same thing.

yes the 2 batteries could be two 6 volt batteries making 12 volts, or 2 twelve volt batteries making 24 volts or one could be an accessory battery.Join the red--positive jumper lead to the positive terminal that supplies the starter motor and the Black jumper lead--negative to the battery terminal - that goes to the body of the vehicle--earth

Rule of thumb---RED cable on POSITIVE Battery connector, BLACK cable on NEGATIVE battery connector and NEVER EVER touch the battery cable conectors together AFTER one end is connected to a battery....!!!!

Yes, as one is probably an accessory battery and will not power the engine systems (starter, ECU, etc).
Connect it to one, if it does not work, try the other. It wont hurt anything if he connects it to the wrong one first... as long as both are 12V systems.
