You'll have to walk or take the subway to find a place less than $150 a night. The Pennsylvania Hotel on 34th Street between 7th and 8th Avenue is less expensive than the Times Square Sheraton or Marriot. The rooms of course aren't as nice, but it's a nice neighborhood next door to Penn Station.

This site is convenient because you can filter search results to match your specific criteria. Try searches for midtown east, madison, lexington, park, fifth ave, and street numbers close to or on 45th street.

It is possible to find affordable rents in NYC, but for the areas you're seeking, you'd be VERY lucky to even find a studio that's less than $1,500 a month.
