when people get to serious about something and stop seeing the humurous and funny aspects of it, do they lose something of their humanity?
does their soul or spirit suffer from it?

There are some things that aren't seen seriously enough. Some things are laughed at inhumanely, which I would say is a bigger threat to our spirits suffering. But I do think some people need to lighten up because being constantly dreary can also cause the spirit to suffer. Life is all about balance.

Yes I think so, and along with losing their sense of humour they seem to lose their sense of empathy...these people become hard, dogmatic and unreachable.
