The various china town buses are cheapest though I would stay clear of Fung Wah. Try Lucky Star bus which is only $20 and has pretty good service. Peter Pan would be the preferred choice from the Port Authority terminal but costs slightly more at about $29 but is more professional. As a side note, you could make the entire trip using public transit for less than $14 via commuter rail through NJ (Trenton) & SEPTA through Philli/ Wilmington ETC and transit buses and so on, but it would take all day.

I would recommend using public transit while in NYC. The reason, parking in the city, specially Manhattan is very tough to find and driving can be a nightmare. Public transit is cheaper than renting a car. Now, if you're wonder how much is will cost you ride the subway in the Big Apple, its $2.25 per ride. However, if you staying in the Big Apple for a week or two, I would recommend purchasing a 7-day or a 14-day unlimited ride MetroCard, which will give you unlimited rides on the city's subway and bus system. The 7-day pass will cost you about $27, while the 14-day pass will cost you $51.50. The unlimited ride MetroCard is huge money saver, specially if you making multiple trips on the subway. I recently took a Greyhound from DC to NYC and paid 20 dollars. It was one of their web deals. Other way round works, too.
Try to get an express bus, so that you don't have to take stops at every house that happens to have been built along the way.

I think all the bus companies lowered their prices after the chinatown buses started pricing so low ($35 roundtrip), but the cheapest is still

Chinatown bus...I don't know the name of the actual company, but there are buses that leave the Port Authority and Chinatown that do incredibly cheap fares.
