When I press the power button on my desktop nothing happens. Checked power cord and power switch on back of PS.

Thinking that if it is a mobo problem that the fans would at least power up; currently nothing happens when button pressed, no fans, drives don't spin up. Nothing.

Anyone know any "easy" ways to check?

Power supply is 500 Watt ThermalTake, Mobo is an MSI K8N Neo; system has been running fine for over two years.

I've worked on computers for years and every time I've had a problem with this, it's always been the power supply.

I've purchased a power supply tester for exactly that purpose. If you don't want to spend the money on the tester, I would purchase a power supply and return it if the new one doesn't work.

Try disconnecting and reconnecting the line that runs from the Mobo to the power switch on your computer. Could just be a small problem with that. Otherwise, it sounds like your power button may be funky, or the line to the mobo might be singed.

you could make sure the plug your plugged into is good. try using another outlet. Also you may want to check and see if the pwer switch is plugged in correctly.

I think the power box in your PC is fused. You would have to replace it.
