Yes they are both voiced by Seth MacFarlane who voices Peter,Stewie, Glenn Quagmire,and Tom Tucker, in Family Guy and Stan Smith in American Dad.
He also has appeared in Star Trek Enterprise as Engineer Ensign Rivers
With ties in nicely with , Patrick Stewart's recurring role as Director Bullock of the CIA in American Dad!

Yes, he also created the show. Brain's voice is his actual normal voice and Stan's is slightly different from how he usually talks.
