Probably James, who supposedly became the first Bishop of Jerusalem. Jesus had four brothers per the "gospels."
"Is he not the carpenter, the son of Mary, and the brother of James and Joses and Judas and Simon? And are not his sisters here with us? And they took offense at him." (Mark 6:3.)

According to the scripture, Jesus had four brothers and several sisters. Of those brothers, two of them would author New Testament books (James and Jude) and James would be the leader of the church in Jerusalem for many years. According to the Jewish historican Josephius, James was killed when the Jewish leaders threw him from the top of the temple. James is also the only one of Jesus' brother and the only other person besides Peter that the Bible list as having a private appearance from Jesus after his resurrection. The other brothers may have seen him at times when he appeared to "everyone".
So while there is no "favorite" brother mentioned in scriputre, James would the most important of the brothers.
