If you look up each one and find the files to delete, you can do it manually, but this is time consuming to go and find each one, and then check to see if it is on your computer.
Utilizing the software, the program has a DAT file, which is a list of spywares to look for as well as the information how to get rid of each instance of the spyware. Using the software is much more expedient as well as more accurate.
A recent study by PC World Magazine showed that there are three programs which, when utilized on the same computer, are the best at getting rid of spyware and Adware problems.
They are:
Spyware Search & Destroy
Adaware SE Personal
and MS Defender

Start by looking for startup programs that you don't recognize. You can find a list using Windows System Information tool. Look for the Startup Programs section under Software Environment. You will see all startup programs and where they are loaded from. Many of them will be loading from Registry keys.
DISCLAIMER FOLLOWS
You can then delete the unwanted startup program files listed as well as the registry entries that reference them. DO NOT attempt unless you are very familiar with the Windows Registry and have recent backups. If you make a mistake in the registry you can destroy your Windows installation. The only reason I have listed this method is because you asked for a removal method without using removal software. Having said that, I recommend using Spybot and CWShredder to start with. Manual removal is less effective and MUCH more frustrating than using the proper tools.

You cannot. You need to have anti spyware software/s first to know if you have spyware and secondly, you will need to know which ones to remove. Without anti spyware, you cannot be certain that your system is spyware free.

That is the only way. You should have at least two spyware protection programs on your computer, along with one virus protection program.
