Well heat rises and cool air sinks so in the corner of the room near the top of the room(if it is a ceiling fan then in the center).
In the corner is best because it can access the entire room. If you place a box fan in the middle of the room you will only be cooling half of the room.
It depends on where you are at, if you are in Arizona the last thing you want id outside air in your house during the summer it is over 110. But if it is the middle of january in alaska the best place is by an open window with room for your body heat to leave the room though the window as well.
I found that if you place a bucket of ice in front of a larger box fan it does a lot better job of keeping me cooler.

EXPLANATION
A fan doesn't actually cool the room unless it's positioned in such a place so as to draw cooler air in. In this respect, if it's colder outside then place the fan next to an open window, simiularly if the room next door is colder then place the fan in the doorway.
If the fan isn't drawing colder air into the room all it will do is circulate the air. The rate of exchange of air in a room is typically 20 to 30 minutes so a fan isn't going to do a great deal unless it's creating enough of a draft that it's actually drawing heat from exposed flesh in the same way that a breeze cools you down on a hot day.
If you're mounting a ceiling fan and it's one that turns in both directions then you want to run the fan so that it pulls the air upwards (usually anti-clockwise unless it's been wired the wrong way round or the blades have been incorrectly mounted). By pulling the air upwards it's lifting the colder air that sinks to the floor, when it's cold the fan wants to push the air downwards so that the warmer air that rises is circulated throughout the room.
If you're going to run a ceiling fan at speed then experimenmt and see which is the most effective and comfortable for you - backwards or forwards.

I would think the best place to keep a fan in the house to get to keep it cool would be..in the center of the room, under the ceiling fan if you have one.

Closer to you. Fans actually heat the room (frictional heat given off by fan motor). While you may get a little mixing to direct the cooler air towards you, most of the coolness will be from the evaporative effect: You sweat, it evaporates and draws heat from you, fan helps remove the moistened and warmed air away from you, bringing cooler and drier air to your skin to repeat the process.

Ceiling fans work the best. The next best thing is a box fan situated in the window. This will keep it really cool at night.
