When my European relatives come and visit, I usually set out a must see list according to the neighborhood in Manhattan. For example on Day 1, I would start out in Battery Park. From there you could visit the Statue of Liberty and Ellis Island. Once you're off the boat, go to Federal Hall and Wall Street. Then you could walk to the west side and see the former site of 9/11 if you like. Still in the neighorhood, you could walk north to City Hall and visit inside the building, quite interesting.
Day 2 - midtown. Start with Empire State Building. Then visit Rock Center, Radio City Music Hall, Macy's, Times Square.
Day 3 - Visit 57th Street and theme restaurants, art galleries, and stores plus the Broadcast museum. Then go west to Lincoln Center. Then go east to Central Park and visit the Zoo. After that walk in Central Park. Keep your eyes open, plenty of celebrities behind sunglasses in the park. Then go to Fifth Avenue and walk to the Metropolitian Museum.
Day 4 - Read the Village Voice, or any local free Manhattan newspaper to see what is going on that day.

It truly depends what your individual intrests are. I can help you out if you preffer artsy stuff. You will most surely want to see a broadway show (even if you do not like musicals, it is a once in a lifetime chance)
If you like dancing (and you are going during the summer) I would go to lincon center. They not only host some of the greatest dance comanies in the world, but they have a free program called midsummernight swing. You can stand in the square and they teach you fun dances. People of all ages go and have a great time.
A large tourist attraction, but something I would highly recomend is the statue of liberty. Especially if you have a family history dating back to that time period it can be facinating, and even if you don't it is still worth seeing.
I would recomend a museum, since NYC is famous for their some museums. Most well known is the Met. Personally I like the museum of naturla history when I was a child. They have dinosour bones, butterfly habitats to walk through, a whole exibit on space, another one on geology, etc.
Other landmark places that are fun to either walk past of briefly enter (just to say you did) include : the empire state building,central park (the zoo is great for kids),Rockafeller center (I HIGHLY recomend this if you are going during the winter: see the tree and go skating) and ground zero.
I hope you have fun. I have been to NYC many times because I live just 1/2 an hour away, and I have foudn one thing to be true every time I go. Even if their is no set itenarary, you will always find something to do. I would recomend chossing a few hours one afternoon and planning to just walk around. Window shop, take pictures, let your individual intrests lead you.

MUST SEE statue of liberty.. it "hits" you once you are there, especially if you had people come over on Ellis island, even if you didn't I didn't get what america was about until I saw ellis islalnd and the statue of liberty.. then just up the street is wall street which everyone should say they have been there
Central Park and Times square give you the flavor of true NYC so you feel like you know it
Soho and the village give you a feel for what it is like to really like in NYC
go to the broadway box office near times square and pick of tickets for cheap just as shown are about to start... be flexible and see anything. We got $300 tickets to MamaMia for $40 but we had to go into the show at that minute and seeing it was the best experience I had in all my trips to NYC, a broadway show

One of the top must see NYC sites is the new Madame Tussaud's Wax Museum at 234 West 42nd Street (between 7th and 8th Avenues). I just went there this past March and it is an unforgettable experience. It is huge and filled with extremely real-looking wax figures, a creeepy (but fun!) Chamber of Horrors interactive exhibit, etc. If you don't get to see too many other museums, you should definitely go here. It's amazing!

Other must-sees are the Bronx Zoo and Coney Island. The Bronx Zoo is the biggest zoo in NYC, and their "Congo" exhibit where you get to view gorillas in their natural habitat is magnificent! If you go on Wednesdays with your family, admission's free!! (although that doesn't count special exhibits like "Congo" and food & drink) but it's very very worth it.

If you have time, swing by a little place called "Manhattan Diner" on the Upper West Side at 2180 Broadway, At 77th St for a meal. Their food is fabulous and cheap. HAVE A GREAT TIME!

with the 3 adults, you can take a visit to the heralded scores strip club in manhattan, only the best there. Catch a yankee/met game if you can, go shopping in manhattan and see some famous landmarks, go to jones beach and have a nice day out there w/ the family, ny is famous for it's food as well, don't be afraid to pig out, and there's lots more but that's more than enough for 4 days
