Its usually said on rap, hip hop forums. They say something that may be interpreted as homosexual, but then they say "no homo" to let others know that it wasnt meant to be interpreted that way.
Like saying "ya Jadakiss always comes hard. (no homo)"
What it really means is that he always spits fire on the track, and hes raw. But dumbasses may see this and interpret in a homosexual connotation, so they say no homo to make sure no-one thinks that way.

'homo' means 'the same' as in homosexual which refers to people who are attracted to and have sex with people of the same sex. it is part of their identity
'no' in this context generally means that something is prohibited, unwanted or unwelcome.
if someone thus says 'no homo' it either means that they are not homosexual themselves and have left out the 't' in 'not', or it means that they have no desire to look outside their box and meet people with a different sexual orientation because they think that homosexuality is a disease, or it means that they are looking for romance and straight romance at that.
