Probably because she is the only one that knows the secret about her painting.
While scholars guess at what the painting means and who is Mona Lisa, she sits smiling and thinking"You will never know and I'll never tell you idiots!"

An interesting question this. I've heard about the theory that she was pregnant, and others, but I recently discovered a new and very interesting theory.
There are some who believe that the Mona Lisa is actually a self-portrait of Leonardo. Don't laugh now! Comparisons have been made, between portrait of Leonardo, and Mona Lisa, and the structure of the faces did show striking similarities. I think an interesting way to see Mona Lisa's smile, is to think of it as Leonardo's smirk, from the fact that he's secretly painting a self-portrait, but no one would know. It is very much in line with his other cryptic ways, such as writing mirror, for example.

In reality, there have been very recent studies that suggest the Mona Lisa was either pregnant or recently had given birth when sitting for the painting. This is all speculation as to why she's actually smiling. Keep in mind this is a portrait. The Mona Lisa was the wife of a well known banker (i believe). Who wants a portrait done that isn't flattering?

I read an article that said when we look at her eyes, we can't focus on her mouth, and the shadows on her cheeks make it look like she is smiling more, but when we focus on her mouth, we see the shadows more clearly and it looks like she isn't smiling as much. It was an interesting article. It went on for awhile, so I know I'm not doing it justice.
I've never thought that it was a self portrait of Leonardo. Most artists put at least a bit of themselves in each portrait they do because of the way artists have left brain short cuts even if we are staring directly at the subject. In fact, all his portraits look a lot like him. The only person I can think of off hand who doesn't do this much would be Velázquez.

actually everyone exagerates this picture too much. Leonardo didn't even like Mona Lisa but had to paint her so he did it with poor effort in spite of her. And he never even finished it.
and to answer you question... its a portrait, smiling is what you're supposed to do.

I'd be more inclined to say she's smiling cause she just got some, and is looking forward to some more later.
In fact, mayhap the wonderous artist had his 'you know what' in his hand and his brush in the other...
That would certainly explain a lot...
