(-: Take a train to Kyoto or Nara -- it's less than an hour's trip.
Seriously, there's Osaka Castle, which is in a nice park. And then there's the eating areas, so be sure to try okonomiyaki, which is an Osaka speciality. We went to Universal Studios, which was nice, but not particularly Japanese (or Osakan). My cousin went to Spa World (public baths) and said it was pretty amazing. Oh, the Hard Rock Cafe near Universal Studios was good, if you like the HRC thing.
Try JNTO (Japan National Tourist Organization) for more info, and I think Osaka City Government has an English tourist site too.

Yahoo! Travel often has some good comments, too. Have fun!

I went to Osaka last year for 2 days. I really enjoyed it there.
Visit the aquarium - it is the best one i have ever been to. There is a whale shark and a sunfish!
The castle looks amazing from the outside and is interesting because there is a lift incorporated on the ouside of it.
The Osaka Museum of Human Rights (Liberty Osaka) is definitely worth a visit.

I have live in Osaka for 7 years.
Everything depends if you want to see the aspect of the Japanese city or the local touristic spot.
In the first case, I strongly recommend you to go to the castle, to enjoy walking in the park and to take a boat going to Minami. It is not Venice, Bruges or Amsterdam but there is lot of charm. When you arrive in Minami, have a walk in America Mura (it is carnival everyday there), go down to Namba, buy local specialities in Dooguya-suji and finish in a restaurant or a club of that area.
If you prefer touristic spots, do not miss the aquarium! It is marvellous. From the aquarium, you can have a look on the Suntory Museum and finish your day in one of the panoramic restaurant of World Trade Center (that one is sake...) on the new port.
In terms of culture, Osaka is not a great area. It is better to go to Kyoto or Nara. But it is a city with a real soul and the best advice I can give you is, as I did, take a map and walk according to your mood. You will find special things every single meter.

Osaka is an amazing city! I recommend you to walk around a bit, see the castle and the aquarium and then....if you truly want to understand what Osaka is all about....go up Osaka Tower and it is pretty amazing once you are on the top floor it will click! You must look at everything on all the floors and work your way up, not down as that won't work! Enjoy!!!

It is 20 min from Osaka to Kyoto by train and 40 min to Kobe, and about 40 min to Nara, if you use Kintetsu line
My advise is to go to see these places first.
In Osaka the best tourist spot are Osaka castle

and Tsuutenkaku

There are many festivals during the summer, but you'd better check them when you arrive.
