Develop a strategy to turn your computer ON and OFF just once. But leave it on for an extended period of time and start an Anti-virus scan or Anti-SpyWare scans while you are not using it. Use Disk Cleanup and Check Disk during these long periods of ON time also. Proper computer maintenance is time consuming, but you don't have to sit and watch. Just plan your schedule around it, and your computer ON/OFF times.

Off when not needed.
More so, to save energy, unplug the computer all together. Saves tons of energy.
And contrary to some belief, this does not hurt the computer unless you start it up again every 2 seconds. But if you shut it down for the night, it's very good practice.

To save energy, and money on your electric bill, off when not needed would be better.
However, I asked a similar question, and someone said that new computers have some sort of energy-efficient way of shutting down, but not completely off.
Additionally, someone else said that they turn it off so that they do not need to replace their fan.

I like to have it off when not in use because: it saves electricity, it saves the lifespan of the fans and electronics in the computer itself, and doesn't suck in all that dust all day long, and it cannot be hacked into from an outside party if it is off.

Depends on how much you use it.
It runs the hard drive to cold boot and that shortens its life. But running it all the time shortens it too. I have always read the 24 hour rule. If you aren't going to be on it tomorrow, shut it off.
FYI: you can schedule virus scans and backup at night so it isn't a total waste.

If you constantly go on your computer then I would leave it on and then every couple of days I would turn it off for a little bit and then turn it back on again.
