I have an interview with another company and currently I'm with a staffing agency.

Warn them ahead of time that you have an appointment on some personal business on (state date) and will have to leave early, this gives them a chance to properly cover for you and makes it sound like no big deal. These things do pop up. This way you are covered if the interview does not work out.

You work for a staffing agency. Are you a temp? Which boss are your talking about? If it is the boss at the company the staffing agency sent you to, tell him/her the truth. S/he knows you are job hunting. It would be polite to give them as much notice as possible.

Your family member called (sister brother grandmother) and can't get in touch with anyone else but you, you have to pick them up right away as they hurt themselves by accident (fell broke a pelvic bone, sliced open an arm trying to cut something never saw across the wrist though up the arm from wrist to innerside of elbow,) and that you need to pick them up as the EMT wont be there for 45 mins.

I would ask for a personal day to take care of some family matters that are very important. This way you are not lieing ...you don't owe them a reason..if you need some time off ask to take it.

Have a friend call and say there's been a family emergency. Say you just puked in the bathroom, say you are having female problems and need to leave

I would just say that you have an appointment. That way you're not lying, but you don't have to divulge the full details, either.

You need the afternoon off for a personal appointment. If he asks what it is, then feel free to b.s. him.
