The best handgun for protection (with accuracy) is the one you have in your hand when you need it most. If you're looking to fill your hand I would suggest a Smith & Wesson K-frame in either .38 Special or .357. My personal preference is a K38 loaded with wadcutters. They punch nice little holes that bleed alot. My next preference is a .41 Magnum revolver loaded with 210 grain lead bullets with about 6 1/2 grains of WW231. This load performs like a .41 Special... if there were a .41 Special. Think of it as a .38 Special with a .41 caliber bullet in a .41 Magnum.

For self-protection you don't need the most powerful or the most accurate. Too much power is actually a hindrance, and many handguns good for hunting will be too much for self-protection. Match-grade accuracy isn't needed, either, since self-defense with a handgun is at or at least near knife-fighting range. Anything from the 38 Special and 9x19 Parabellum on up to the 45 ACP and 45 (Long) Colt will do nicely, except that the 44 Remington Magnum (apologies to Dirty Harry) with full-power loads will overpenetrate and actually be less effective than lighter loads in most defensive situations.
There are tons of good choices, and your specific situation will determine what's best for you. If you live in the middle of nowhere your choice will be different than if you live in a city. Whether you feel the need to carry all day or just want something for the home will make a big difference. Your past experience and willingness/ability to practice a lot will also be a determinant.

The words "powerful" and "accurate" don't necessarily define a handgun used for self protection. The most powerful would probably be the new S & W .460, in addition to killing the intruder, you might just kill 3 others in the next block who are in the line of fire. The most accurate would be some high dollar single shot target pistol, but the accuracy is limited by the skill of the shooter.
For self defense purposes, accurate is "How good can you point your finger?" Since most gunfights in the home are limited to less than 20 feet, the issue is can you point your finger at the target.
Any hand gun larger than .380 with frangible or hollow point bullets that you can stand the recoil and hit the target the 2nd time would be a good choice.

For personal/home defense you really don't need anything more powerful than a .357 Magnum. Nowadays, most handguns are as accurate as the shooter so that is a moot point.

You didn't say if you preferred revolvers or auto-loaders so...

Revolver: a S & W, Ruger or Taurus .357 Magnum

Pistol: Glock .9mm, .40 S & W or .357 Sig.

For power any of the custom made pistols that occasionally turn up chambered for the 50 BMG.

For commercial production it could the Encore based pistols from SSK Industries they come in calibers such as 458WIN, 475 JDJ, 470 Nitro Express and the 50 Alaskan.

Accuracy on the other hand has a finicky nature. The accuracy of a gun can vary from day to day due to changing weather. So its pretty much impossible to declare that one chambering is both the most power full and the most accurate.

Boy, that's tuff to say. There are a plethora of great guns out there. It all comes down to what you are willing to spend, and what feels good to you. I have a friend who swears by Glock 23 .40 caliber. He shoots it very accurately, and it is a good weapon. I can't shoot it for shat though. My hands are too big for it. I prefer the Springfield XD .40 Caliber. I like the safeties on it and it feels good in my hand. As far as caliber, there is a never ending debate about which round you should buy for "Knock down power". If you can hit what you are aiming at it doesn't really matter. It's only as lethal a your aim anyway. Other good names in guns include, but are not limited to, Kimber, HK, Sig, and Styre. Good luck picking one. I've always though of buying a gun as the men's version of women's shoe shopping. Finding the right one is half the fun.

Accuracy, power, and self-protection are not necessarily all going to be found in one gun. If you are looking for a concealed carry gun, I would suggest a small frame revolver, .38 or .357 caliber. If you are looking for home protection, either a larger frame revolver or a pump-action shotgun. First and foremost, though, is a gun safety course and target practice.

I do recall seeing a 45/70 revolver at sportsmans outpost...........that would be a serious recoil tho........ at close range that should stop a bison......
