DNA can be changed, this is called a mutation and it happens in cancerous cells. However, in order for DNA to change like Spidy's did, it would have to happen in the entire body which is highly unlikely. Most of the time mutations occur on select cells and this is why most forms of cancer are localized to certain tissues like the liver for example. A change in DNA occurs in one cell at a time, so in order for the changes in spidy's DNA to take place they would have to take place at an extreemly fast rate or whatever was causing the changes would have to occur in many different cells at once. It is highly unlikely.

Yes DNA can be altered, although as previous posters have mentioned, it has to occur in every cell of the body. Some types of viruses can be engineered to do this, although it would be difficult and time consuming.
The other issue would be figuring out exactly what changes to make. DNA is mostly gibberish in between genes. Think of it as trying to read a book where the pages are filled with letters, but there is only one word on every one to two pages. On top of that, it's in a language no one understands. In order to change the meaning of the book not only do you have to find the right words to change, you have to change them into the correct new word in every copy of the book (i.e. cell) on the planet (body).

Of course. Only when all the DNA in every single cell in the body undergo the SAME change all at once. It is just not practical. I suggest changing the DNA right at the start, when the "person" is still a single cell embryo. That might work. Ethical issues are another problem altogether.
