Everyone of us has a specific calling in God's ministry.
One person maybe called to work in children's ministry.
Another one maybe called to stuff missionary letters.
Another one might feel God leading him to become a pastor.
God calls us to serve him in various ways.
Its a challenge sometimes to figure out where God wants us.
God speaks to each of us in our hearts where we should serve.

The sayin "This is my calling" or "God is calling me", means that you have found what it is that the Lord wants you to do with you life, or what it is that God wants you to do. Romans 11:29 "For the gifts and calling of God [are] without repentance."
God has a special plan for everyone in life. Some have bigger things that God has called them to do, but those who lead ordinary lives also are called of God; maybe not to do any huge thing, but the gifts that the Lord has given them are not to be wasted and if they convert even just one soul, then their lives are just as important as those who travel to other countries and have large ministries.

This is my calling means that you found out what God has planned for you to do in life; that you found your destiny and purpose. When they say God is calling me they often say that means God wants you to do something.

God gives your soul certain gifts, talents and abilities when you're in the womb. when you set these gifts, talents and abilities to work for God, they are called "anointings". so a preacher would be anointed to teach
usually when someone says "God is calling me to do this", they mean they feel this is what they feel they are set apart to do. some giftings and talents are less obvious (example, you would make a good poet, but you've never given it a chance)
and some are very obvious (a pastor should be a good socializer and teacher and be insightful)

If God has called you to do something then you have dilligently seeked His will for your life. You have prayed, and meditated over what it is that He wants you to do. And then it is getting that answer to your prayer.
