It has many, many meanings, depending on the context. For example "Los" can mean "fortune", "batch", or "lottery ticket", but "Was ist los" means "What's the matter", and "Los gehts!" means "Off we go!". Consult a dictionary like dict.cc : http://www.dict.cc/?s=los

Was ist den los? Loose translation = what is wrong? More of a technical translation = what is then missing? You are missing something, something is not right, when something is wrong - would you agree? Was ist den los is a saying that has a different meaning, it's a colloquialism. (sp? I probably murdered that.)

Was ist los. Loosely, "what's wrong", "what's happening"
