I suppose it means that once you get your heart's desire your life is fulfilled and you won;t have anything to aspire to anymore.

To me the first part of it is obvious. You get what you want.
The second part refers to the “chase”. The pursuit, working hard to obtain something. Once you’ve obtained that what you want the most, what is left? If one were to actually discover the meaning of life, what would the point of living be? It would be a tragedy to no longer have a goal, no longer have a reason to work, to strive to obtain more. The tragedy would be having no reason to go on.

That's just another way of saying "You're damned if you do and you're damned if you don't" because the human heart is in a constant state of searching for something new, something more, something better, etc.
Once a goal is reached one often asks "Is that all there is?" or "Now what?" How tragic it is for people to put so much effort into obtaining something (or someone) that they don't take the time to really enjoy what is already set before them.
Discontentment, dissatisfaction and disillusionment are always somewhere lurking in the corners of our minds but we have the innate ability to change this. Why don't we? Why don't we?

The first is the tragedy of longing...the second is the tragedy of your attaining your goal and it's not the thing you really needed...like winning the lottery and still being miserable.

I think it means we spend an absurd amount of time wishing for things we want and missing out on what we do have, and then when we get what we want, the reality of it does not live up to the fantasy.
