592 Fifth Avenue is at 48th Street/5th Av.
There is no one train you could take that would get you to 66 John St directly there but this will get you close.
Walk over to the 49th Street/7th Av station on the N, R, W and take any downtown train on stop Times Square-42nd St. Change (using the staircases at the rear of the platform on the N,R, W platform) for the downtown #2 or #3 and take either one to Fulton Street (William St) . Make sure you use the William St exit. Walk down one block to John St. 66 John is probably just off the corner of William and John.
(Note: You can also walk a short distance more to the Broadway/50th St and take the downtown #1 one stop to Times Square. The downtown #2 & #3 trains are right across the platform there).
