The folks who set up the catagories would know, but I think I can offer a decent idea as to why they're two catagories, rather than a single one.
"Religion & Spirituality" — think of this one as covering various beliefs and philosophies generally thought of as being current.
"Mythology & Folklore" — this one is usually considered to cover various beliefs and philosphies which are no longer currrent.
(There are other possible explanations, of course, but here I just want to attempt a nice, simple answer without offering offense to anyone or starting an argument.)
If you're fortunate, you might get an answer from someone who actually had a hand in setting up the catagories and get an "official" answer.

The categorization has been done by Yahoo!Answers, and they only will be able to explain. People actually does not know in which category they should put their question and it is all mixed up. Mythology and Folklore section is supposed to be about past folklore items. It is still interesting and you can search the question of your choice.
The Religion and Spirituality section is a big ocean and always there are more than 4000 questions unresolved and it is difficult to identify the questions of your interest. Many Mythology & Folklore questions are put in Religion & Spirituality.
Yahoo!Answers should clear the confusion by merging both catagory and devide it into different new catagories on some correct basis...

Supposedly, Religion and Spirituality is "real" and Mythology and Folklore are considered to be "unreal".
I try to stay away from the Religion and Spirituality category because you always end up being attacked when you answer those questions.
I prefer Mythology and Folklore...and to me, it's a lot more real than religion and all its cohorts.

Because judeo christian beleivers can't see the simple logic that what they consider myths and folklore are nothing more than the devoutly beleived relegions of other people. They call them stories and the bible fact.LOL.
