I think I would have to go with Chuck Berry. I see the best band as one that would have to be at least somewhat revolutionary. Why are the Beatles widely considered the best? Because of the way they changed music forever. I see the Doors and Kiss and Motley Crue and aside from being incredibly hedonistic I don't really see their contribution. Maybe Kiss for being the glam rock pioneers, but the way I see it where are they without Chuck Berry. The man brought the electric guitar into the forefront of music. Oh, and he totally rocked.
I know you could say he is a person and the question is "band" but he had a band and most bands are driven by a central member anyway.

The Doors, Tom Petty & the Heartbreakers, The Allman Brothers Band, Jimi Hendrix Experience (considered British because two of the members was from England, but Jimi was American and he counts as 5!), The 13th Floor Elevators, The Black Crowes (17 years old, not new are they? well, they're newer than the others), Canned Heat, Cinderella, Grateful Dead, Dr. Hook and the Sloppy Seconds...then, the best American band of all, CCR!!!
Of course, there are plenty I didn't mention. Those are a few of my favorites though.
