It depends on the kinds of painkillers you are taking either non-narcotic or narcotic painkillers.
> Non-narcotics. Overdosing or taking too much painkiller acetaminophen, best known as Tylenol can poison the liver.You won't die instantly or while sleeping. You might be taken to hospital and depending on the severity of the extent of the damage there will be very high chance of having death on the end. Overdosing or taking too much different painkillers -- such as aspirin, ibuprofen, naproxen or ketoprofen -- called NSAIDs, or nonsteroidal anti-inflammatory drugs will result to severe side effects including stomach bleeding and kidney problems.Extensive damage incurred could result to high possibility of dying at the end as well.
>Narcotics. Overdosing with narcotics painkillers such as morphine and pethidine and other opioid painkillers which have muscle relaxation effect could end up slowing down the muscles of vital organs. As the result, there will be respiratory depression, slowed heart functions,and all the systems will be extremely slow. So you could instantly at a very short time or while you are sleeping.

Normally the respiratory control center in the medulla monitors the PCO2 levels (technically they monitor the H+ ion concentration), and the PO2 levels in your blood. When the H+ levels go above a certain concentration, and the O2 levels fall below a certain partial pressure, the respiratory control center stimulates the diaphragm via the phrenic nerve, causing inspiration.
Narcotics like morphine, heroin, etc. disrupt this mechanism, and the body becomes desensitized to decreasing oxygen availability. The brain cells require a steady supply of oxygen, so when the respiratory system is depressed, brain cells die . . . which means you die.

Killing yourself is wrong so don't do it.
Opiates/muscle relaxant mixtures are propably the least painful, the opiate creates a sense of euphoria and the muscle relaxants eventually halt your breathing.
Downing a bottle of Tylenol. This causes massive liver damage, if someone tries to overdose with this the pain would most likely be intense and prolonged.

depends upon how much you take, but usually your body would slowly start to shut down, starting with your respiratory system, then everyother system would shut down. You would pass out before you died, to answer your question
