Chances are the driver for your sound card is not compatible with Windows Vista. This is not an uncommon problem since many software and hardware vendors have not released drivers which support Windows Vista. Go to the sound card manufacturer's website and see if they have a newer driver which support Windows Vista. Or you can look for a driver at (http://www.radarsync.com/vista/) which is a repository of Vista ready drivers.

make sure your sound drivers are up to date. Also, look up in the sound properties and see if anything is muted, but my guess is the drivers.
