It means "You don't want to go to the store with us (us meaning girls)"

Don’t you want to go to the store with us (girls)?
OR
Do you not want to go to the store with us (girls)?

It is “us (girls)” because the Spanish word “nosotras” is applied to girls. The word “nosotros” is for boys.

Although literally it would mean: Don't you want to go to the store with us (girls)? IF it is a question...that it seem to be.

It seems better to translate it:

Would you like to go to the store with us (girls)?

It means "You don't want to go to the store with us?"
<< the 'us' is refering to girls only because nosotrAS was used instead of nosotrOS

Without question mark - you don't want to go to the shop with us. (perhaps the girls wanted a girly outing . . .)
With question mark - don't you want to go to the shop with us? (this is more of an invitation.)
