I was in NYC last yr for a over a week so I was able to spread things out more, but the one thing I loved and would totally recommend is doing a harbor tour. They have a 2hour tourney and then a 3hr tour. The 3hr tour takes you right around Manhattan Island, you get to see a lot of stuff, and get as close to the Statue of Liberty that you can without stepping onto the island. I found it well worth it and learnt a lot about NYC that I wouldn't have other wise.
Also take a cab ride, those guys have some great stories to tell.

*Smirks* You should see me! New York's greatest attraction.
Just kidding. If you have a night to party, I'd suggest checking out Coyote Ugly on 1st Ave and E. 9th Street.
It's a really fun, crazy bar. It usually makes for a memorable night, and they've got $2 cans of Pabst Blue Ribbon which hold costs down, and make you feel like you're in college again.

Walk around Times Square. That place is amazing! And, everyone else is right when they say take a rode in a cab somewhere. The ride is better than a roller coaster at six flags. I have never experienced driving like this before. Let's just hope you get a good one! Also, eat at Carmine's (sp?). It is the bet Italian food you'll ever eat! And have fun. I'm jealous, I wanna go!!! :((
