If you want to ask for permission to do something:

me das permiso xxxx ? = will you allow me xxxx
If you want to walk between people, instead of saying "excuse me", you have to say: "permiso" or "con permiso"

may I do it = puedo hacerlo ?

as the above sample, asking directly to a person, you don't need to specify sex (he or her).

asr for her permission = "pídele permiso a ella"
