I don't know if you could say they were the "best" (there's always somebody better), but the most influential, hands down, were Jimmy Hendrix and Jimmy Page.
Both awesome guitarists. I was in rehab with Jimi Hendrix's back-up bass player, an old guy who knew Jimi Hendrix in Vietnam. Seems Jimi was an 82nd Airborne trooper.

Early rock pioneer: Chet Atkins--he inspired Jimi and Eddie.
Artistically: Jimi Hendrix and Stevie Ray Vaughn (both deceased, but honored sincerely--just the same.)
Dynamically (to this day) Eddie Van Halen.
The fact that NO "Alternative" rockers appear listed tells one something, doesn't it?

Talk about a can of worms. I don't know that anyone can say who the "best is" but here are some of my rock guitar gods: Jimmy Paige, Pete Townsend, John McLaughlin, Mark Knopfler, Eric Clapton. If you want to branch into jazz and blues add Mark Whitfield, Wes Montgomery, Stevie Ray Vaughan, Mark Stern.

Terry Kath. Hendrix once told Chicago their guitarist (Kath) was better than himself. Coming from Hendrix, that's saying a lot.
