Ruger New Vaquero

It depends on which .45 caliber cartridge you are talking about and if you want it for hunting, target shooting or self-defense.

.45 Long Colt

For hunting, get a Ruger Blackhawk in .45 LC. You can handload to the lower levels of .454 Casull.

For defense, an S&W M-625 or M-625 Mountain Gun would be a good choice. A 3" barrel if you can find it, or a 4" barrel would be good. However, they won't handle the heavy loads a Blackhawk witl take.

Colt Peacemakers and their clones are fair for hunting, but the fixed sights limit it as a hunting gun. A 4 5/8" SAA still makes a helluva fine defensive sixgun.

.45 ACP

Now we are talking games and self defense.

Model 1917 revolvers from either S&W or Colt will make a fine defensive sidearm. You can still find them at fairly modest prices, but the prices are climbing.

For serious target and competition shooting, an S&W M-625 .45 ACP is hard to beat.

It depends on what you are thinking of using it for. If you are thinking of cowboy shoots I would go with a Black Hawk .45 Long Colt, because it is a single action and in most cowboy shoots a single action is required. If I where using it for hunting and target I would buy a Ruger Red Hawk .454 Casull. You can shoot a .45 long colt out of the .454casull. It will be cheaper and more comfortable for targeting and a .454 casull for hunting. I disagree with the guy that says you can shoot a .454 Casull out of a .45 Long Colt. That is trouble,it isn't built to withstand the pressure.

the best 45 is one made like the old 1911.just get a good brand and not some knock off.the knock off will not perform like a good brand.try to always use factory ammo in a auto weapon. reloads tend to jam and not eject propperly
