Wow - I wrote the above, but you've received so many differing answers you will never be able to choose! I'm going to explain it further.....
You can actually say it two ways - I wrote the one I would choose above. Here's how to tell the difference:
DeberÃa haber dos personas aquà hoy.
There should be 2 people here today (We're expecting them, we don't know if they will show up or not, but that's the plan. This version has a little bit of the "unknown" suggested by the verb ending "Ãa.")
Debe haber dos personas aquà hoy.
There should be 2 people here today (We're expecting them, and they should be here. This version is a little more matter-of-fact.)
The difference is so very subtle however, we tend to just prefer one or the other. Whichever one you choose will be perfectly understood.

Either "Deben de haber dos personas aqui hoy" or "Estan supuesto haber dos personas aqui hoy". You can also start either phrase with the word "Hoy" rather than at the end.
